√ Product Description:
The Thawing Kit consists of Thawing Solution (TS), Dilution Solution (OS), and Washing Solution (WS). These are HEPES-buffered liquids containing human serum albumin and gentamicin sulfate.
TS and OS contain trehalose as a non-permeable cryoprotectant, while WS does not contain any cryoprotectant.
√ Scope of Application:
Intended for the thawing of human oocytes, pronuclear-stage embryos, cleavage-stage embryos, and lastocysts .
√ Instructions for Use:
1. Thawing must be performed at 37℃(on a heating block).
2. Prepare a culture dish with 1 ml of Thawing Solution (TS) at least 30 minutes before thawing, and preheat to 37°C. Prepare dishes with 50 loll droplets of Dilution Solution (OS) and Washing Solution (WS) at room temperature.
3. Transfer the cryopreserved oocytes/embryos from the storage container to a liquid nitrogen holding vessel near the microscope and laminar flow hood.
4. Immerse the cryopreservation straw containing the oocytes/embryos in the preheated Thawing Solution (TS) at 37℃. Position the oocytes/embryos in the solution for 1 minute.
5. Transfer the oocytes/embryos to Dilution Solution (OS) for 3 minutes, then wash in Washing Solution (WS) for 5 minutes. Transfer to fresh WS for an additional 3 minutes of washing.
6. Continue with routine laboratory procedures for further culture or transfer.
