Sperm Freezing:
Collect semen after 2-3 days of sexual abstinence and allow to liquefy at room temperature or 37℃ for 15-30 minutes. Warm an equal volume of Sperm Freezing
Solution to room temperature or 37℃. Transfer liquefied semen to a sterile 15mL conical centrifuge tube. Add the Sperm Freezing Solution dropwise to achieve a 1:1 ratio with the semen sample, mixing gently after each drop to avoid osmotic shock and bubble formation. Close the centrifuge tube tightly and invert 20 times to mix thoroughly. Aliquot the semen mixture into cryovials or straws labeled with the patient's ID, leaving space at the closed end to prevent expansion during freezing. Seal the cryovials or straws according to the manufacturer's instructions. Float the samples in a room temperature water bath for 10 minutes, then equilibrate in a 2℃ refrigerator for 90 minutes. Place the samples in the vapor phase of liquid nitrogen for 30 minutes. Quickly transfer the cryovials or straws
into liquid nitrogen and store at -196℃.
Sperm Thawing:
Remove cryovials or straws from liquid nitrogen (-196℃) and place in a water bath at 35±2℃ for 30 seconds. Dry the cryovials or straws with clean paper, carefully open them, and transfer the semen mixture to a sterile test tube. Slowly add an equal volume of gamete buffer dropwise, mixing gently to avoid shock and bubble formation. Finally, purify the sperm using a sperm density gradient separation medium.
